Dato' Supiah Bt. Mohd Yusof started her career with the Public Service Department in 1975 and had then served in various capacities in the Department. Dato' Supiah was also a Member holding various positions in the Majlis Agama Islam Negeri Pulau Pinang.

In the year 1996, Dato' Supiah served in the Bahagian Penyelerasan Penyertaan Bumiputera Pulau Pinang, Unit Penyelarasan in the Prime Minister’s Department as the Assistant Director. She then served as the Deputy State Financial Officer for the State of Penang in year 2001. In year 2004, she was the Deputy Secretary in the State Secretariat Department, before being transferred as State Financial Officer of Penang in April 2006.
